My laundry sink set up. Water won't drain from sink and water is also backing up to vent line. The vent is open, and have a air admittance valve on laundry sink. Nothing drains...do I have this set up right? Is it more likely a blockage issue?

Is this your lowest drain? I am picturing a basement, perhaps with no floor drain. How long since your septic was pumped?
