Depending on age of the system or when the home was built, some homes will have dampers to close off or reduce air flow to certain registers. Most homes will have a registers where you can close it off some to restrict air flow and allow more air to flow to other rooms. FYI, registers usually mean air from the air handler, grills are the returns to the air handler.
At least in Florida, to prevent stagnation of air flow for closed off rooms such as bedrooms, jump vents are installed which all it is a grill in the ceiling with duct work to another ceiling grill on the other side of the door, usually the bedroom hallway. Rooms that seem cold with the door closed is a lack of air flow from the register. To prove it with the door closed, feel for air moving under the closed door.
There are several ways to measure air flow with a tool to "balance" the system. Plenty of info on line on how to's.
This is a wall register and the lever on the right is adjustable.
