Installing a Vespin II and the unifit screw holes are going to line up directly in the grout line on one side. The other is in the center of the tile. Is this an issue?

If it is then I will have to use a different toilet that does not use the adapter and have to be bolted to the floor.

If you can drill the hole, this is going to work. The hole thru the tile can be oversized, but the hole below the tile should be pilot hole sized.

Having the unifit screw holes lining up differently on your tile may not be a major issue, but it's a good idea to ensure a secure installation to avoid any future problems. If it concerns you, consider consulting a professional or the toilet manufacturer for guidance.
