There are several ways to anchor things into concrete, but I believe the easiest way is to use lead ferrels and sheet metal screws. Use the flange as a template and drill holes for the ferrels with a rotary hammer/drill. Then drive stainless steel sheet metal screws into the ferrels. Get the largest size screws that will fit the flange and match the ferrel size to the screw size. I don't have a clue why you have a 6" drain, but since it is new, perhaps you can contact the builder and find out.
