Yes you will need to take up concrete.
Everything needs to be vented.
A basement bathroom install is a big job and probably not a good first plumbing project. You may want to do the "labor" part of the job--removing the concrete and dirt necessary and opening the walls needed to get the vent to the roof--and hire a plumber to run the drainage, vent and water lines.
