Most new toilets flush fairly quickly, so noise will be noticed in the line, unless there is better sound damping on the pipe and wall below.
Like you mention, putting board over concrete doesn't really do much.
It needs to be isolated.
Many times with concrete, there will put attach metal strips to the wall and ceiling that hold the drywall away and allow for vibration dissipation.
